Timing gene expression programs is essential to form and maintain neuronal diversity. Using human organoid models we develop, we dissect the mechanisms that time the unfolding of developmental genetic programs to generate spatial patterns of neuronal subtypes, ultimately forming distinct sensory-motor circuits along the spinal cord. We further explore how disruptions in the timing of intracellular transport may perturb gene expression programs to induce motor neuron disease.
